L-ARGININE AMIDINASE - Introduction
The quarternary structure of native rat liver arginase has been described by Hirsch-Kolb, H & Greenberg, D.M.," Molecular characterisitics of rat liver arginase", J. Biol Chem., 243, 6123-6129 and Baranczyk-Kuzma A., Porembska Z. & Mochnacka, I., Acta Biochim Polon, 23, 151-163. EDTA treatment dissociated the enzyme into inactive subunits of 30,000 daltons each. Addition of Mn2+ ions restored the activity and caused reassociation of subunits to the natve form of 120,000 daltons.

L-ARGININE AMIDINASE - Physiological effects
Studies have found that up-regulation of arginase has a great impact on vascular function and is one of the causes of vascular dysfunction, but the mechanism of up-regulation is still unclear and may include the participation of multiple mediators. The activation of arginase by cysteine-iron is dose-dependent, accompanied by the formation of • OH free radicals. Interestingly, when the production of • OH is inhibited by salicylic acid derivatives and superoxide dismutase, the activity of arginase is also correspondingly inhibited, suggesting that • OH free radicals may be involved in the activation of arginase. Cytokines such as IL-4 and IL-13 can up-regulate the expression of arginase I, and supplement of exogenous arginine can restore the ability of IL-4 treated macrophages to produce NO. H2O2 can also up-regulate arginase expression, thereby inhibiting NO-mediated endothelium-dependent coronary artery dilatation. At present, there are few researches on the signaling pathway regulating arginase expression in vascular cells.
